Evaluación de primers en la identificación del gen PEPT1 en diferentes tejidos de juveniles híbridos de tilapia red florida

In the present investigation primer sets for identification in the PepT1 gene in different tissues of juvenile hybrid tilapia Red Florida were used. The goal was to find, within four primer pairs designed by bioinformatics, the pair of specific primers in the amplification of gene PepT1. The sample processing began with ethanol fixation in 96% of tissue juvenile tilapia temperature under two conditions: Ambient and deep freezing at -18 ° C. The DNA extraction protocol used was that of Genomic DNA Mini Kit PureLink® (Catalog Numbers K1820-01, K1820-02, K1821-04) Invitrogen®. For PCR amplification of different regions belonging to PepT1 gene, various modifications of the programming performed thermocycler. The amplicons were analyzed by agarose gel electrophoresis. Primer set was more effectively Fw 11 - 6bRev as a single band in electrophoresis was visualized, the other had double band, additionally the condition ideal holding temperature was room for its better observation gel. This study will help in later investigations nutritional tilapia, allowing lower costs and shorter time required for analysis by use of this first specific also measuring gene expression evidence PepT1 nutrient assimilation linked thereto.
