Abstract
The objective of this study was to evaluate the HCM effect (mango peel flour, Mangifera indica L) on abdominal obesity, lipid profile and morphometry of the intestinal epithelium. The experiment was carried out in the Agricultural Laboratories of the Technical University of Manabí in 28 male CD1 mice, weighing 25 to 30 grams. The mice were randomly distributed and received the food corresponding to the following treatments Conventional (T1), conventional + 4% HCM (T2), hyper fat (T3) and hyper fat + 4% HCM (T4) diets for 16 consecutive weeks. After the experimental period, the mice were previously sacrificed overnight fasting, with an intraperitoneal (IP) injection of a mixture of xylazine (25 mg / Kg) and ketamine (150 mg / Kg), blood samples were obtained by exanguination (1 to 1.5 ml), which are used for the analysis of the lipid profile and blood glucose by spectrophotometry. Abdominal fat, liver and heart were extracted from each experimental unit and the relative weight of these organs was calculated. In addition, jejunum and duodenal samples will be taken, these samples were preserved in 10% formalin of general staining of hematoxylin and eosin in paraffin sections. The results were analyzed by Analysis of Variance (ANOVA), Completely Random Design (DCA) with 2x2 factorial arrangement and the Tukey mean comparison test (p <0.05). The results obtained were: higher food consumption in T1, while the highest caloric intake, abdominal fat deposition and final weight were detected in the treatments that received a hyper fat diet (p <0.05). The relative weight of the liver is not different between treatments (p> 0.05), while the relative weight of the heart is lower in T3 in relation to the remaining treatments (p <0.05). In addition, the serum concentration of glucose, total cholesterol, triacylglycerol, LDL and the HDL / LDL index was not different between treatments (p> 0.05). Additionally, the serum concentration of glucose, total cholesterol, triacylglycerol, LDL and the HDL / LDL index was not different between treatments (p> 0.05). However, a higher concentration of HDL in T4 (p <0.05) was reported; the CT / HDL ratio (atherosclerotic index) tended to be lower (0.072) in diets supplemented with HCM (T2 and T4). Modification in intestinal morphometry (length, width and depth of crypts) is not modified. In conclusion, these results suggest that HCM could influence the lipid profile of mice, without modifying somatic parameters. El objetivo de este estudio fue evaluar el efecto HCM (harina de cáscara de mango, Mangifera indica L) sobre la obesidad abdominal, perfil lipídico y morfometría del epitelio intestinal. El experimento se realizó en los Laboratorios Agropecuarios de la Universidad Técnica de Manabí en 28 ratones macho CD1, de 25 a 30 gr de peso. Los ratones fueron distribuidos aleatoriamente y recibieron la alimentación correspondiente a los siguientes tratamientos Dietas convencional (T1), convencional + 4% HCM (T2), hipergrasa (T3) y dieta hipergrasa + 4% HCM (T4) durante 16 semanas consecutivas. Culminado el periodo experimental los ratones se sacrificaron previo ayuno nocturno, con una inyección intraperitoneal (IP) de una mezcla de xilazine (25 mg/Kg) y ketamina (150 mg/Kg), las muestras de sangre se obtuvieron mediante exanguinación (1 a 1,5 ml), que se utilizó para el análisis del perfil lipídico y glucemia mediante espectrofotometría. Se extrajo la grasa abdominal, hígado y corazón de cada unidad experimental y se calculó el peso relativo de estos órganos. Adicionalmente, se tomaron muestras de yeyuno y duodeno, estas muestras se conservaron en formol al 10% tinción general de hematoxilina y eosina en cortes de parafina Los resultados se analizaron mediante el Análisis de Varianza (ANOVA), Diseño Completamente al Azar (DCA) con arreglo factorial 2x2 y la prueba de comparación de medias de Tukey (p < 0,05). Los resultados obtenidos fueron: mayor consumo de alimento en T1, mientras la mayor ingesta calórica, deposición de grasa abdominal y peso final se observó en los tratamientos que recibieron una dieta hipergrasa (p < 0,05). El peso relativo del hígado no fue diferente entre tratamientos (p > 0,05), mientras el peso relativo del corazón fue menor en T3 en relación a los tratamientos restantes (p < 0,05). Adicionalmente, la concentración sérica de glucosa, colesterol total, triacilglicerol, LDL y el índice HDL/LDL no fue diferente entre tratamientos (p > 0,05) Sin embargo, se reportó mayor concentración de HDL en T4 (p< 0,05); la relación CT/HDL (índice aterosclerótico) tendió a ser menor (0,072) en las dietas suplementadas con HCM (T2 y T4). No se observó modificación en la morfometría intestinal (largo, ancho y profundidad de las criptas). En conclusión, estos resultados sugieren que la HCM podría incidir en el perfil lipídico de los ratones, sin modificar parámetros somáticos.
